Malezija in Borneo : Potopisno predavanje na 15. Dia FEST-u v Ljubljani

Abstract: Z Malezijo sta imeli s Tino neporavnane račune, saj je obisk leta 2020 preprečil nanometrski velikan znan pod imenom SARS-CoV-2. Letos spomladi pa je končno poletela v Kuala Lumpur, kjer se je tritedenska avantura začela. Po zaužitju nekaj čarov ikonske arhitekture jo je mahnila v višavje s precej bolj prijetno klimo in neskončnimi plantažami čaja, med katerimi kraljujejo rastlinjaki z jagodami in drugimi kulturami, ki jim tropska vlaga in vročina ne ustrezata. Pot nadaljuje proti severovzhodu dežele na rajske otoke Perhentian. Načrtovala je sicer oddih, a so jo lepote koralnih grebenov, pisane ribice, morske želve, skati in celo (vegetarijanski) morski psi vedno znova vabili v svoj objem pod vodo. Večino časa tako preživi s potapljaško masko na glavi. Vrne se na celino in poleti v svetovljanski Georgetown, kjer se znajde v »mali Kitajski« in se zabava ob iskanju uličnih grafitov. A vročina in vlaga sta tukaj še bolj ubijalski kot v prestolnici, zato se umakne na Penang Hill, kjer nekoliko lažje zadiha. Sledi let v Kuching in raziskovanje Bornea, kjer jo navduši na novo odprt Muzej bornejskih kultur, še bolj pa stik z naravo. Obišče divje orangutane in nacionalni park Bako, ker se sreča z makaki, opicami rilčarji, srebrnolistnimi opicami, bornejskimi divjimi prašiči, pa tudi s strupeno zeleno suličarko. Po siloviti tropski nevihti komaj posuši obleke in gojzarje, ko je že čas za vrnitev na celino, ogled zgodovinskega mesta Melaka, zadnje nakupe in let proti Evropi...

Fenolni potencial izbranih vin s podaljšano maceracijo sorte "Rebula"

Abstract: V diplomski nalogi smo analizirali 30 vzorcev vin sorte grozdja Vitis vinifera L. cv. 'Rebula' iz Vipavske doline in Goriških brd. Vina smo kupili na trgu, in sicer v specializiranih lokalnih prodajalnah vina, spletnih prodajalnah ali pri vinarjih. Osemindvajset (28) vzorcev vina je bilo pridelanih s postopkom podaljšane maceracije drozge grozdja. Dve vini pridelanih brez maceracije, smo dodali v nabor vzorcev zaradi primerjave fizikalno-kemijskih parametrov z ostalimi vzorci. V vinskih vzorcih smo izmerili naslednje standardne fizikalno-kemijske parametre: skupni in prosti žveplov dioksid, skupne titrabilne kisline, pH, hlapne kisline z destilacijsko metodo, izbrane organske kisline, sladkorje (glukozo + fruktozo), etanol in barvo vina s spektofotometrom - absorbanco pri 420 nm (A420) in barvne parametre v CIE Lab sistemu. Izmerili smo tudi skupne polifenole po spektrofotometrijski metodi Folin-Ciocalteu in izbrane posamezne polifenole (hidroksibenzojske kisline, nezaestrene hidroksicimetne kisline, procianidine) s tekočinsko kromatografijo visoke ločljivosti, sklopljeno z UV detetktorjem (HPLC-UV). Želeli smo določiti polifenolni potencial vin sorte 'Rebula', pridelanih s postopkom podaljšane maceracije drozge grozdja in nato določiti morebiten vpliv časa maceracije na količino polifenolov v vinu. Kemijske analize so pokazale veliko variabilnost v količini skupnih polifenolov, ki pa jih nismo uspeli povezati s časom maceracije. Pomanjkljivi podatki o času in drugih pogojih maceracije drozge grozdja nam tega niso dopuščali. Podatki nakazujejo trend večjih koncentracij skupnih polifenolov vina pri vinih, pridelanih s postopkom podaljšane maceracije drozge grozdja. V določenih vinih smo izmerili tudi preko 1000 mg/L (kot galna kislina) skupnih polifenolov. Med posameznimi polifenoli je bilo v vinih največ galne kisline (tudi do 90 mg/L), procianidina B1 (do 52 mg/L) in katehina (do 18 mg/L). Instrumentalno določena barva filtriranega vina (A420 in parametri CIE Lab) je povezana s količino skupnih polifenolov po metodi Folin-Ciocalteu. Parametri barve dolgo maceriranih vin se močno razlikujejo od vin brez maceracije oziroma od vin, pridelanih s krajšim časom maceracije, zato gre za dobre 'markerje' tovrstnih vin. Opazili smo, da kombinacija običajnega parametra barve belih vin A420 nm s parametri CIE Lab sistema omogoča podrobnejšo analizo barve vina.

Abstract: The economic analysis showed that the MAST project is justified and represents added value for all users, the financial feasibility of carrying out the MAST project as such was confirmed. The set business model of the MAST project was proved to be sustainable in the long run, as the project from 2022 onwards shows considerable self-sustainability. Not only during its implementation but also at its finish the project may be estimated low-risk also for its possible future developments, as it clearly promotes an innovative idea and features a clear business model based on a professional team in capacity to face all the challenges and risks.

POLYMORPHISMS IN GENES FOR ENDOTHELIN 1, ENDOTHELIN RECEPTORS AND NITRIC OXIDE SYNTHASE 3 IN PATIENTS WITH DIABETIC RETINOPATHY AND DIABETES MELLITUS TYPE 2 : DISSERTATION

Abstract: The major causes of Diabetes Mellitus Type 2 (T2DM) are multi-factorial consequences of complex interactions between environmental, social and genetic factors. We investigated the genetic risk factors in Slovene patients with T2DM on a model of microvascular complication – Diabetic Retinopathy (DR). Retrospective case-control study includes a T2DM Slovene population with clinical risk factors for T2DM and DR. Only some of the candidate genes with selected single nucleotid polymorphisms (SNPs) were included: (EDN1 (rs5370, rs3087459, rs1476046), EDNRA (rs5335, rs1801708), EDNRB (rs10507875, rs4885493), NOS3 (rs869109213). By genotyping with either real-time polymerase chain reaction or standard polymerase chain reaction (PCR) we successfully identified the contribution of variable number of tandem repeats rs869109213 in DR progression (Proliferative Diabetic Retinopathy (PDR)) in Slovene patients with T2DM. The joint effect of individual genotypes of rs10507875 in EDNRB and rs869109213 in NOS3 on DR onset (DR) and DR progression (PDR) was demonstrated as well. The joint effect of the two polymorphisms on DR onset (DR) and DR progression (PDR) was greater than the individual effect of each polymorphism separately in the analyzed genetic models. Despite genetic research contributions in DR, linkage studies, and Genome-wide association studies the identification of susceptible loci through candidate gene approaches still remains in its early stages. The frequent approach with an ultimate focus on SNP associations with phenotype is likely to underestimate the roles of genetics in human diseases by disregarding not only the joint effect of multiple loci but the complex interaction network between them. By identifying polymorphisms in genetic disorders in a more systematic way, we will be able to deepen our understanding of the regulatory mechanisms and disease etiology which should lead to a more effective development of mechanism-based therapies as well.
