Monitoring of the particles above the unpaved road by lidar technique

Abstract: Basic concepts of laser-based technique LIDAR (LIght Detection and Ranging) are discussed in this article as well as advantages and disadvantages. Presented are measurements of relative particulate concentrations above an unpaved road which were performed in a rural area near the Iowa City, Iowa, USA. The LIDAR system used in these measurements is a small, scanning LIDAR that uses elastic backscattering to obtain information on the amount of atmospheric aerosols. In the elastic LIDAR, light scattered back towards the LIDAR system from molecules and particles in the atmosphere is collected by a telescope and is detected with a photodiode. A Big Sky Laser model CFR-200 Nd:YAG laser operating at 1.064 microns is used to generate the LIDAR's outgoing signal. The laser is attached directly to the top of a 26 cm, f/10 Cassegrian telescope. A series of pulses are summed to make a single scan. A series of scans is used to build up a two- dimensional map of relative atmospheric aerosol concentrations. With a maximum range of about 6-8 km, a range resolution of 2.5 m, and a time resolution of 30 s, the LIDAR is capable of very detailed mapping of aerosols.

1,8-dihydroxy naphthalene (DHN) - melanin confers tolerance to cadmium in isolates of melanised dark septate endophytes

Abstract: The contribution of 1,8-dihydroxy naphthalene (DHN) melanin to cadmium (Cd) tolerance in two dark septate endophytes (DSE) of the genus Cadophora with different melanin content was investigated in vitro. The DSE isolate Cad#148 with higher melanin content showed higher tolerance to Cd than the less melanised Cad#149. Melanin synthesis was significantly reduced by Cd in both isolates with uninhibited melanin synthesis, in a dosedependent manner. Inhibition of melanin synthesis by tricyclazole reduced the relative growth of Cad#148 exposed to Cd and did not affect Cad#149. Cd accumulation was not altered by tricyclazole in the two isolates, but it increased catalase and reduced glutathione reductase activity in more melanised Cad#148, indicating higher stress levels. In contrast, in Cad#149 the enzyme activity was less affected by tricyclazole, indicating a more pronounced role of melanin-independent Cd tolerance mechanisms. Cd ligand environment in fungal mycelia was analysed by extended EXAFS (X-ray absorption fine structure). It revealed that Cd was mainly bound to O- and S-ligands, including hydroxyl, carboxyl, phosphate and thiol groups. A similar proportion of S- and Oligands (~35% and ~65%) were found in both isolates with uninhibited melanin synthesis. Among O-ligands two types with Cd-O-C- and Cd-O-P- coordination were identified. Tricyclazole altered Cd-O- ligand environment in both fungal isolates by reducing the proportion of Cd-O-C- and increasing the proportion of Cd-O-P coordination. DHN-melanin, among other tolerance mechanisms, significantly contributes to Cd tolerance in more melanised DSE fungi by immobilising Cd to hydroxyl groups and maintaining the integrity of the fungal cell wall.

Arbuscular mycorrhizal fungi alter Hg root uptake and ligand environment as studied by X-ray absorption fine structure

Abstract: Mercury (Hg) – plant – fungal interactions are only poorly studied. Hg speciation and ligand environment in maize roots inoculated with arbuscular mycorrhizal (AM) fungi were investigated in order to better understand the role of AM in Hg soil to root transfer. The maize plants were grown in Hg polluted substrate (50 mg g1 as dissolved HgCl2) and inoculated with AM fungi originating from: a) highly Hg polluted environment of a former Hg smelting site in Idrija, Slovenia, (Glomus sp. – sample AmI), and b) non-polluted environment (commercial AM inoculum Symbivit1 – sample AmC). Hg speciation and ligand environment in maize roots was studied by Hg-L3 XANES and EXAFS with emphasis on XAS methodology – modelling and fitting the XAFS spectra to extract in a reliable way as much information on Hg coordination as possible. The AmI plants developed more arbuscules and less vesicles than the AmC plants, and also accumulated more Hg in the roots. A clear difference in Hg coordination between the AM (AmC & AmI) and the control (ConC & ConI) plants is recognized in Hg L3-edge EXAFS analysis: in the ConC & ConI maize roots 73–80% of Hg is attached between two sulphur atoms at the distance of 2.34 Å. The remaining ligand is nitrogen at 2.04 Å. In AmI & AmC roots another Hg-S attachment encompassing four thiol groups at the S-distance of 2.50 Å are identified, accounting for 21–26%. AM fungi can modify Hg ligand environment in plant roots, thus playing an important role in biogeochemical cycling of Hg in terrestrial ecosystems.

ŠTUDIJA VKLJUČITVE ZUNANJIH DOBAVITELJEV V ENOTNI POSLOVNI INFORMACIJSKI SISTEM PODJETJA PIPISTREL : MAGISTRSKO DELO

Abstract: Način komuniciranja med naročnikom in dobaviteljem vpliva na uspešnost in učinkovitost poslovanja podjetja. Osnovni namen magistrske naloge je prikazati pomen komuniciranja med naročnikom in dobaviteljem. Odraz dobre komunikacije je tudi razumevanje naročil in izpolnjevanje pričakovanj naročnika. Cilj predlaganega dela je predstaviti poslovno funkcijo nabave, opredeliti ključne nabavne procese, pripraviti načrt za izvedbo izboljšav in ga implementirati v informacijski sistem podjetja. Obravnavali smo možnosti vključevanja dobaviteljev v enotni sistem ERP (Enterprise resource planning) in predlagali rešitve za izboljšanje komunikacije, preko katerih bo podjetje učinkoviteje sledilo naročilom. S preučitvijo toka informacij med podjetji in na osnovi obstoječega načina komunikacije smo predlagali elektronsko obliko sodelovanja, ki zunanje dobavitelje vključuje v enotni sistem ERP podjetja Pipistrel. Na konkretnem primeru dobavitelja smo opisali simulacijo predlaganega sodelovanja z aplikacijo Ominet, da pri naročilih in dobavi ne bi prihajalo do napak. Predlagano sodelovanje naj bi bistveno prispevalo k izboljšanju kakovosti dobavljenega blaga, kontrole kakovosti procesov in posledično končnih izdelkov podjetja. Vključitev v enotni sistem ERP dobaviteljev dodatno bistveno ne obremenjuje, naročnik in dobavitelj pa s tem pridobita na kakovosti. Opisali bomo testno vključitev uporabnika in doseganje pričakovanih rezultatov. Upravljanje z aplikacijo Ominet je enostavno, uporabnik ima boljši pregled nad dokumentacijo, naročila so bolj ažurna in profesionalno izvedena. Magistrsko delo vsebuje navodila za uporabo aplikacije, prikazan je tudi postopek, ki dobavitelju omogoča izvajanje operacij v naročnikovem sistemu ERP preko oddaljene povezave.
