Aerosol complex refractive index retrieval in the Paris urban area and its forested surroundings during the ACROSS field campaign : variability and constraint for direct radiative effect estimation in regional models

Opis: The complex refractive index (CRI) is one of the key parameter driving aerosol spectral optical properties and direct radiative effects (DRE). Its value and spectral variation under different conditions, such as anthropogenic− and biogenic−dominated environments and anthropogenic−biogenic mixing situations, remains not fully understood. As a consequence, oversimplified representations of aerosol optical properties are generally used in climate models. Therefore, measurements of aerosol CRI in different environments and their inclusion in models are needed. The field observations from the ACROSS campaign, performed in June-July 2022 in the Ile de France region, are used in this study to deepen the knowledge of aerosol optical properties, aiming to improve the aerosol representation in the CHIMERE model and provide the best constraint for DRE simulations. Measurements obtained both at the Paris city center and the Rambouilllet rural forest sites during ACROSS are considered, in order to explore the CRI variability from anthropogenic−dominated to biogenic−dominated environments, including anthropogenic−biogenic mixing situations. The CRI retrievals at seven different wavelengths, performed by combining the Mie theory with optical and size distribution measurements, are representative of different atmospheric conditions, aerosol loadings as well as type and chemical compositions. In fact, the June-July 2022 period was characterized by highly diversified weather conditions: 1) two strong heatwaves, promoting SOA build-up and favoring the export of the Paris pollution plume towards the forest site; 2) Saharan dust events transported from the upper atmosphere to the ground; 3) biomass burning episode; 4) periods with reduced anthropogenic influence. The CRI retrievals under these different conditions and their link to particulate chemical composition is investigated. Hence, the CRI dataset presented here constitutes a unique dataset from which models can benefit to validate and constrain simulations and DRE estimations, under both urban and biogenic emissions influence. These data, in conjunction with those from the aircraft observations during ACROSS, are used to initialize and perform sensitivity studies on the aerosol DRE, using the CHIMERE−WRF coupled model, the OPTSIM model for the aerosol optical properties and the Rapid Radiative Transfer Model for GCMs (RRTMG).Keywords: Complex refractive index, direct radiative effect, aerosol mixing, urban, forest

Composition and diversity analysis of the lung microbiome in patients with suspected ventilator-associated pneumonia

Opis: Background: Ventilator-associated pneumonia (VAP) is associated with high morbidity and health care costs, yet diagnosis remains a challenge. Analysis of airway microbiota by amplicon sequencing provides a possible solution, as pneumonia is characterised by a disruption of the microbiome. However, studies evaluating the diagnostic capabilities of microbiome analysis are limited, with a lack of alignment on possible biomarkers. Using bronchoalveolar lavage fluid (BALF) from ventilated adult patients suspected of VAP, we aimed to explore how key characteristics of the microbiome differ between patients with positive and negative BALF cultures and whether any differences could have a clinically relevant role. Methods: BALF from patients suspected of VAP was analysed using 16s rRNA sequencing in order to: (1) differentiate between patients with and without a positive culture; (2) determine if there was any association between microbiome diversity and local inflammatory response; and (3) correctly identify pathogens detected by conventional culture. Results: Thirty-seven of 90 ICU patients with suspected VAP had positive cultures. Patients with a positive culture had significant microbiome dysbiosis with reduced alpha diversity. However, gross compositional variance was not strongly associated with culture positivity (AUROCC range 0.66–0.71). Patients with a positive culture had a significantly higher relative abundance of pathogenic bacteria compared to those without [0.45 (IQR 0.10–0.84), 0.02 (IQR 0.004–0.09), respectively], and an increased interleukin (IL)-1β was associated with reduced species evenness (rs = − 0.33, p < 0.01) and increased pathogenic bacteria presence (rs = 0.28, p = 0.013). Untargeted 16s rRNA pathogen detection was limited by false positives, while the use of pathogen-specific relative abundance thresholds showed better diagnostic accuracy (AUROCC range 0.89–0.998). Conclusion: Patients with positive BALF culture had increased dysbiosis and genus dominance. An increased caspase-1-dependent IL-1b expression was associated with a reduced species evenness and increased pathogenic bacterial presence, providing a possible causal link between microbiome dysbiosis and lung injury development in VAP. However, measures of diversity were an unreliable predictor of culture positivity and 16s sequencing used agnostically could not usefully identify pathogens; this could be overcome if pathogen-specific relative abundance thresholds are used.

Untargeted molecular analysis of exhaled breath as a diagnostic test for ventilator-associated lower respiratory tract infections (BreathDx)

Opis: Patients suspected of ventilator-associated lower respiratory tract infections (VA-LRTIs) commonly receive broad-spectrum antimicrobial therapy unnecessarily. We tested whether exhaled breath analysis can discriminate between patients suspected of VA-LRTI with confirmed infection, from patients with negative cultures. Breath from 108 patients suspected of VA-LRTI was analysed by gas chromatography-mass spectrometry. The breath test had a sensitivity of 98% at a specificity of 49%, confirmed with a second analytical method. The breath test had a negative predictive value of 96% and excluded pneumonia in half of the patients with negative cultures. Trial registration number: UKCRN ID number 19086, registered May 2015.

Effects of polyethylene terephthalate (PET) microplastics and acid rain on physiology and growth of Lepidium sativum

Opis: This study evaluated the chronic toxicity (30 days) of different sizes of polyethylene terephthalate (PET) microplastics (60e3000 mm) provided alone or in combination with acid rain, on garden cress (Lepidium sativum). Both biometrical and physiological traits have been evaluated: i) percentage inhibition of seed germination, plant height, leaf number and fresh biomass production; ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione production); iii) impairment in photosynthetic machinery in term of pigments production; iv) aminolevulinic acid and proline production. Results highlighted that different sizes of PET, alone or in combination with acid rain, are able to negatively affect both biometrical and physiological plant traits. In particular, the lower size of microplastics is able to negatively affect growth and development, as well as to trigger the oxidative burst. Regarding the pigments production, PET coupled with acid rain, induced a higher production of Chl-b, and an inhibition of aminolevulinic acid.

Short-term physiological and biometrical responses of Lepidium sativum seedlings exposed to PET-made microplastics and acid rain

Opis: Plastics enter in terrestrial natural system primarily by agricultural purposes, while acid rain is the result of anthropogenic activities. The synergistic effects of microplastics and acid rain on plant growth are not known. In this study, different sizes of polyethylene terephthalate (PET) and acid rain are tested on Lepidium sativum, in two separate experimental sets. In the first one we treated plants only with PET, in the second one we used PET and acid rain together. In both experimentations we analyzed: i) plant biometrical parameters (shoot height, leaf number, percentage inhibition of seed germination, fresh biomass), and ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione). Results carried out from our experiments highlighted that different sizes of polyethylene terephthalate are able to affect plant growth and physiological responses, with or without acid rain supplied during acute toxicity (6 days). Spectroscopic insights into the electrochemical mechanism of rechargeable calcium/sulfur batteries

Opis: Calcium batteries represent a promising alternative to lithium metal systems. The combination of the low redox potential and low cost and the energy-dense calcium anode (2073 mAh/cm3, similar to 2044 mAh/cm3 for Li) with appropriate low-cost cathode materials such as sulfur could produce a game-changing technology in several fields of applications. In this work, we present the reversible activity of a proof-of-concept Ca/S battery at room temperature, characterized by a surprising medium-term cycling stability with low polarization, promoted by the use of a simple positive electrode made of sulfur supported on an activated carbon cloth scaffold, and a state-of-the-art fluorinated alkoxyborate-based electrolyte. Insights into the electrochemical mechanism governing the chemistry of the Ca/S system were obtained for the first time by combining X-ray photoelectron spectroscopy and X-ray absorption spectroscopy. The mechanism implies the formation of different types of soluble polysulfide species during both charge and discharge at room temperature, and the formation of solid CaS at the end of discharge. The reversible electrochemical activity is proven by the reformation of elemental sulfur at the end of the following charge. These promising results open the way to the comprehension of emerging Ca/S systems, which may represent a valid alternative to Mg/S and Li/S batteries.
