Silver nanoparticles enter the tree stem faster through leaves than through roots

Opis: A major environmental pollution problem is the release into the atmosphere of particulate matter, including nanoparticles (NPs), which causes serious hazards to human and ecosystem health, particularly in urban areas. However, knowledge about the uptake, translocation and accumulation of NPs in plant tissues is almost completely lacking. The uptake of silver nanoparticles (Ag-NPs) and their transport and accumulation in the leaves, stems and roots of three different tree species, downy oak (Quercus pubescens Willd.), Scots pine (Pinus sylvestris L.) and black poplar (Populus nigra L.), were assessed. In the experiment, Ag- NPs were supplied separately to the leaves (via spraying, the foliar treatment) and roots (via watering, the root treatment) of the three species. Uptake, transport and accumulation of Ag were investigated through spectroscopy. The concentration of Ag in the stem was higher in the foliar than in the root treatment, and in poplar more than in oak and pine. Foliar treatment with Ag-NPs reduced aboveground biomass and stem length in poplars, but not in oaks or pines. Species-specific signals of oxidative stress were observed; foliar treatment of oak caused the accumulation of H2O2 in leaves, and both foliar and root treatments of poplar led to increased O2− in leaves. Ag-NPs affected leaf and root bacteria and fungi; in the case of leaves, foliar treatment reduced bacterial populations in oak and poplar and fungi populations in pine, and in the case of roots, root treatment reduced bacteria and increased fungi in poplar. Species-specific mechanisms of interaction, transport, allocation and storage of NPs in trees were found. We demonstrated definitively that NPs enter into the tree stem through leaves faster than through roots in all of the investigated tree species.

A multielement analysis of Cu induced changes in the mineral profilesof Cu sensitive and tolerant populations of Silene paradoxa L.

Opis: tThis work investigates the Cu induced changes in element profiles in contrasting ecotypes of Silene para-doxa L. A metallicolous copper tolerant population and a non-metallicolous sensitive population weregrown in hydroponics and exposed to different CuSO4treatments. Shoot and root concentrations of Ca,Cu, Fe, K, Mg, Mn, Mo, Na, P, S and Zn were evaluated through ICP-OES.Results indicated that increasing the environmental Cu concentration had a population dependenteffect on element profiles, shoot-to-root ratios and correlations among the elements. Generally, in thetolerant population Cu treatment induced a higher element accumulation in roots and had minimaleffects on the shoot element profile, thus resulting in a progressively decreasing shoot-to-root ratio foreach element. In the sensitive population element concentrations in root and shoot were much moreaffected and without a consistent trend. Copper treatment also affected the correlations between theelements, both in roots and shoots of the two populations, but more so in the sensitive population thanin the tolerant one. Thus, Cu exposure strongly disturbed element homeostasis in the sensitive population,but barely or not in the tolerant one, probably mainly due to a higher capacity to maintain proper rootfunctioning under Cu exposure in the latter. Differences in element profiles were also observed in theabsence of toxic Cu exposure. These differences may reflect divergent population-specific adaptations todifferential nutrient availability levels prevailing in the populations’ natural environments. There is noevidence of inherent side-effects of the Cu tolerance mechanism operating in the tolerant population.

Fibroblast protein profile analysis highlights the role of oxidative stress and vitamin K recycling in the pathogenesis of pseudoxanthoma elasticum.

Opis: Pseudoxanthoma elasticum (PXE) is a genetic disorder associated to mutations in the ABCC6 gene; however, the pathogenetic mechanisms leading to elastic fibre calcifications and to clinical manifestations are still unknown. Dermal fibroblasts, directly involved in the production of the extracellular milieu, have been isolated from healthy subjects and from patients affected by PXE, cultured in vitro and characterized for their ability to produce reactive oxygen species, for structural and functional properties of their cell membranes, for changes in their protein profile. Data demonstrate that oxidative stress has profound and endurable consequences on PXE fibroblast phenotype being responsible for: reduced levels of global DNA methylation, increased amount of carbonylated proteins and of lipid peroxidation products, altered structural properties of cell membranes, modified protein expression. Data shed new light on the pathogenetic pathways in PXE, by identifying a network of proteins affecting elastic fibre calcification through inefficient vitamin K recycling, and highlight the role of differentially expressed proteins as targets for validating the efficacy of future therapeutic strategies aiming to delay and/or revert the pathologic phenotype of PXE fibroblasts. Moreover, data open new perspectives for investigating PXE-like phenotypes in the absence of ABCC6 mutations.

Use of reactive tracers to determine ambient OH radical concentrations: Application within the indoor environment

Opis: The hydroxyl radical (OH) plays a key role in determining indoor air quality. However, its highly reactive nature and low concentration indoors impede direct analysis. This paper describes the techniques used to indirectly quantify indoor OH, including the development of a new method based on the instantaneous release of chemical tracers into the air. This method was used to detect ambient OH in two indoor seminar rooms following tracer detection by gas chromatography- mass spectrometry (GCMS). The results from these tests add to the small number of experiments that have measured indoor OH which are discussed with regard to future directions within air quality research.
