1. Primerjava ročnega in avtomatskega sistema spremljanja membran za reverzno osmozoIbrahim Mujezinović, 2023, diplomsko delo Opis: Ta diplomska naloga raziskuje prehod iz ročnega na avtomatsko spremljanja membran reverzne osmoze. V nalogi je opisan koncept reverzne osmoze in podobnih filtracijskih tehnologij, kot tudi pomembnost spremljanja stanja membran med postopkom filtracije. Prav tako je predstavljena vloga senzorjev pri avtomatskem spremljanju delovanja membran.
V nalogi so opisane različne metode za spremljanje stanja membran, s poudarkom na senzorjih. Razloženo je, kako senzorji delujejo in kako zbirajo relevantne podatke o delovanju membran. Prav tako je opisan način, kako omogočajo avtomatsko spremljanje.
Predstavljena je tudi primerjava med ročnim in avtomatskim spremljanjem membran reverzne osmoze. Razložene so prednosti avtomatskega spremljanja, kot so hitrejše odkrivanje težav, zmanjšanje človeških napak in izboljšanje celotne učinkovitosti postopka filtracije.
Na koncu naloge je izvedena analiza, ki preučuje uporabo membran reverzne osmoze s senzorji in brez njih. The conformational plasticity of the selectivity filter methionines controls the in-cell Cu(I) uptake through the CTR1 transporterPavel Janoš, Jana Aupič, Sharon Ruthstein , Alessandra Magistrato, 2022, izvirni znanstveni članek Opis: Copper is a trace element vital to many cellular functions. Yet its abnormal levels are toxic to cells, provoking a variety of severe diseases. The high affinity copper transporter 1 (CTR1), being the main in-cell copper [Cu(I)] entry route, tightly regulates its cellular uptake via a still elusive mechanism. Here, all-atoms simulations unlock the molecular terms of Cu(I) transport in eukaryotes disclosing that the two methionine (Met) triads, forming the selectivity filter, play an unprecedented dual role both enabling selective Cu(I) transport and regulating its uptake rate thanks to an intimate coupling between the conformational plasticity of their bulky side chains and the number of bound Cu(I) ions. Namely, the Met residues act as a gate reducing the Cu(I) import rate when two ions simultaneously bind to CTR1. This may represent an elegant autoregulatory mechanism through which CTR1 protects the cells from excessively high, and hence toxic, in-cell Cu(I) levels. Overall, our outcomes resolve fundamental questions in CTR1 biology and open new windows of opportunity to tackle diseases associated with an imbalanced copper uptake. Antibodies against four independent tumor antigens were isolated by panning a nanobody synthetic library. Such antibodies enabled flow-cytometry-based sorting of distinct cell sub-populations from UM PDXs and to analyze their genomic features. The complexity and specificity of the biochemical and genomic biomarker combinations mirrored the UM tumor polyclonality. The data showed that MUC18 is highly and universally displayed at the surface of UM cells with different genetic background and consequently represents a reliable pan-biomarker for their identification and purification. In contrast, the other three biomarkers were detected in very variable combinations in UM PDX cells. Biotinylated Vanadium and Chromium Sulfide Nanoparticles as Probes for Colocalization of Membrane ProteinsAlexandre Loukanov, Saim Emin, 2016, izvirni znanstveni članek Opis: ABSTRACT We report the microemulsion synthesis of vanadium and chromium sulfide nanoparticles (NPs) and their biological application as nanoprobes for colocalization of membrane proteins. Spherical V2S3 and Cr2S3 NPs were prepared in reverse microemulsion droplets, as nanoreactors, obtained by the surfactant sodium bis(2-ethylhexyl) sulfosuccinate (AOT) in nonpolar organic phase (heptane). Electron microscopic data indicated that the size distribution of the nanoparticles was uniform with an average diameter between 3 4 5 nm. The prepared hydrophobic nanocrystals were transferred in aqueous phase by surface cap exchange of AOT with biotindihydrolipoic ligands. This substitution allows the nanoparticles solubility in aqueous solutions and confer their bioactivity. In addition, we report the conjugation procedure between a-Lipoic acid (LA) and biotin (abbreviated as biotin-LA). The biotin-LA structure was characterized by 1D and 2D NMR spectroscopy. The biotinylated vanadium and chromium sulfide nanoparticles were tested as probes for colocalization of glutamate receptors on sodium-dodecyl-sulfate-digested replica prepared from rat hippocampus. The method suggests their high labeling efficiency for study of membrane biological macromolecules.
