Nanobodies: towards rational design of immune-reagents

Abstract: Antibodies are irreplaceable reagents in both research and clinical practice. Despite their relevance, the structural complexity of conventional mono- and polyclonal antibodies (IgG) has always been a limit for their engineering towards reagents optimized for specific applications, such as in vivo diagnostics and therapy. Furthermore, their isolation is time consuming, their production expensive, and their functionalization results often in heterogeneous macromolecule populations. These drawbacks promoted the search for both innovative antibody isolation strategies and alternative scaffolds. In vitro panning of pre-immune collections of recombinant antibody fragments allows for the simple and fast recovery of binders. Since they did not undergo somatic maturation, their affinity for targets can be insufficient but on the other hand they can be rapidly mutated by standard molecular biology techniques to generate second-generation antibodies among which to identify clones with improved characteristics. Both stochastic and rational methods have been proposed for the optimization process. Random mutagenesis followed by panning at stringent conditions has been successful used to select binders with improved physical characteristics. Rational methods try to identify in silico key residues involved in the regulation of specific antibody features, such as stability or binding affinity. The accuracy of these methods usually depends on the calculation resources. In this perspective, smaller molecules can be analyzed “better” than larger because of their restricted number of residues. Nanobodies small dimensions have been long appreciated since enable better tissue penetration, shorter clearance time, higher yields. Now it becomes evident that this characteristic makes them also optimal objects for modeling.

The non-dominant hand perseveration and movement in SZJ locative constructions

Abstract: In sign languages, signers habitually encode the relations between locative arguments with a complex predicate consisting of several independent morphemes, as shown by Pfau and Aboh (2012) for Sign Language of the Netherlands. In this study, I examine perseverations and movements of the non-dominant hand (H2) in Slovenian Sign Language (SZJ) locative constructions. In SZJ, the H2may be persevered after producing the two-handed Ground in locative constructions. This is shown by the data collected from seven first language SZJ informants, using a Picture Description Task. The referential location as well as the orientation and the handshape of this perseveration may change at the sign-boundary when the one-handed Figure has just been articulated and the one-handed predicate is about to be signed. Before this sign-boundary, the handshape of the persevered H2 refers to the Ground – and is therefore a Ground classifier. After that boundary, the handshape of the persevered H2 refers to the part of the Ground that is relevant for localizing the Figure – and is therefore an axial part classifier that projects aMeasure Phrase.

Influence of Gas Dynamic Virtual Nozzle Geometry on Micro-Jet Characteristics

Abstract: In this paper we present a numerical study investigating the effects of nozzle geometry on stability, shape and flow characteristics of micron-sized liquid jets, produced by injection molded gas dynamic virtual nozzles (GDVNs) operating in vacuum. The jet characteristics are described as a function of (i) capillary-to-orifice distance, (ii) nozzle outlet orifice diameter, and (iii) liquid feeding capillary angle. An experimentally verified numerical model of GDVN with laminar two-phase Newtonian compressible flow, based on finite volume method and volume of fluid interface tracking, is used to assess the changes. The study is performed for two sets of liquid flow rates while keeping the gas flow rate constant. It is observed that for each value of capillary-to-orifice distance and nozzle outlet diameter there is a minimum liquid flow rate below which the jet is unstable. We find that the nozzle outlet diameter has the biggest influence on the jet diameter, length and velocity, while liquid capillary angle has no observable effect on jet characteristic. Varying capillary-to-orifice distance does not affect the flow field around micro-jet. It is found that the liquid and the gas interaction near the meniscus primarily affect the jet stability and shape
