13. Laser ablation-single-particle-inductively coupled plasma mass spectrometry as a multimodality bioimaging tool in nano-based omicsDino Metarapi, Johannes Teun van Elteren, Martin Šala, Katarina Vogel-Mikuš, Iztok Arčon, Vid Simon Šelih, Mitja Kolar, Samo B. Hočevar, 2021, original scientific article Abstract: Advanced data processing and visualization techniques were applied to laser ablation-single particleinductively
coupled plasma mass spectrometry (LA-sp-ICPMS) data obtained from imaging of roots of
sunflower (Helianthus annuus L.) which were exposed to ionic silver (Ag+). Highly multiplexed images with
tissue level resolution were generated, showing the uptake and transformation of Ag+ to silver
nanoparticles (AgNPs) in whole root cross-sections, and revealing that the size of biosynthesized AgNPs is
predisposed by the reducing power of particular root compartments. Various Ag+ and AgNP visualization
strategies are shown with special emphasis on spatially resolved information associated with the number
and spherical-equivalent size of individual AgNPs in user-selected root cross-sectional regions, including
statistical analysis of the AgNP distribution. Normally, this multimodality information can only be obtained
by combining the results of techniques from several complementary platforms. Keywords: Laser ablation, Ag XANES, bioimaging Published in RUNG: 03.06.2021; Views: 2093; Downloads: 50 Link to full text This document has many files! More... |
14. Preparation of porous [alpha]-Fe[sub]2O[sub]3 thin films for efficient photoelectrocatalytic degradation of basic blue 41 dyeManel Machreki, Takwa Chouki, Mitja Martelanc, Lorena Butinar, Branka Mozetič Vodopivec, Saim Emin, 2021, original scientific article Abstract: A novel method was developed for the preparation of porous hematite (α-Fe2O3) thin films. First, a solution containing iron precursor was spin-coated on fluorine-doped tin oxide substrate and later short heat-treated at 750 °C. The prepared α-Fe2O3 thin films were applied as dual-function catalyst in photoelectrochemical (PEC) water oxidation and textile dye degradation studies. For the first time, α-Fe2O3 thin films were used in efficient PEC degradation of a textile dye (Basic Blue 41 – B41) using in-situ generated reactive chlorine species. In comparison with photocatalytic and electrocatalytic approaches, the PEC technique allows faster degradation of B41 dye at an applied bias potential of 1.5 V versus reversible hydrogen electrode and under visible light illumination. In the presence of Cl− using the PEC approach the degradation of B41 reaches 99.8%. High-performance liquid chromatography coupled with UV–VIS system confirmed the degradation of B41 dye using PEC. Gas-chromatography coupled to mass spectrometry was used to study the by-products obtained during PEC degradation. Chemical oxygen demand analyses confirmed that the mineralization level of B41 is in the order of 68%. The α-Fe2O3 films developed in this study give a higher level of PEC degradation efficiency compared to other iron oxide-based systems. 15. Temporal and spatial patterns of zinc and iron accumulation during barley (Hordeum vulgare L.) grain development. Journal of agricultural and food chemistry.Amelie Detterbeck, Paula Pongrac, Daniel Persson, Katarina Vogel-Mikuš, Mitja Kelemen, Primož Vaupetič, Primož Pelicon, Iztok Arčon, Søren Husted, Jan Kofod Shjoerring, Stephan Clemens, 2020, original scientific article Abstract: Breeding and engineering of biofortified crops will benefit from a better understanding of bottlenecks controlling
micronutrient loading within the seeds. However, few studies have addressed the changes in micronutrient concentrations,
localization, and speciation occurring over time. Therefore, we studied spatial patterns of zinc and iron accumulation during grain
development in two barley lines with contrasting grain zinc concentrations. Microparticle-induced-X-ray emission and laser ablationinductively
coupled plasma mass spectrometry were used to determine tissue-specific accumulation of zinc, iron, phosphorus, and
sulfur. Differences in zinc accumulation between the lines were most evident in the endosperm and aleurone. A gradual decrease in
zinc concentrations from the aleurone to the underlying endosperm was observed, while iron and phosphorus concentrations
decreased sharply. Iron co-localized with phosphorus in the aleurone, whereas zinc co-localized with sulfur in the sub-aleurone. We
hypothesize that differences in grain zinc are largely explained by the endosperm storage capacity. Engineering attempts should be
targeted accordingly. Keywords: barley (Hordeum vulgare L.), biofortification, grain development, grain loading, LA-ICP-MS, μ-PIXE Published in RUNG: 20.10.2020; Views: 2763; Downloads: 0 This document has many files! More... |
20. Cadmium associates with oxalate in calcium oxalate crystals and competes with calcium for translocation to stems in the cadmium bioindicator Gomphrena clausseniiPaula Pongrac, Tania Serra, Hiram Castillo Michel, Katarina Vogel-Mikuš, Iztok Arčon, Mitja Klemen, Boštjan Jencic, Anja Kavčič, Mina T. Villafort Carvalho, Mark G. M. Aarts, 2018, original scientific article Abstract: Cadmium (Cd) was shown to co-localise with calcium (Ca) in oxalate crystals in the stems and leaves of
Cd tolerant Gomphrena claussenii, but Cd binding remained unresolved. Using synchrotron radiation
X-ray absorption near edge spectroscopy we demonstrate that in oxalate crystals of hydroponically
grown G. claussenii the vast majority of Cd is bound to oxygen ligands in oxalate crystals (488%;
Cd–O–C coordination) and the remaining Cd is bound to sulphur ligands (Cd–S–C coordination). Cadmium
binding to oxalate does not depend on the amount of Ca supplied or from which organs the crystals
originate (stems and mature leaves). By contrast, roots contain no oxalate crystals and therein Cd is
bound predominantly by S ligands. The potential to remove Cd by extraction of Cd-rich oxalate crystals
from plant material should be tested in phytoextraction or phytomining strategies. Keywords: Cd XANES, Cd oxalate, Gomphrena claussenii Published in RUNG: 06.09.2018; Views: 3345; Downloads: 0 This document has many files! More... |
