262. Cortisol fetal fluids and newborn outcome in term pregnancy small-sized purebred dogs.Barbara Bolis, Tanja Peric, Alessandro Rota, Massimo Faustini, Maria Cristina Veronesi, 2016, published scientific conference contribution abstract Abstract: In order to provide further information about canine perinatology, and because of the scarce
knowledge about fetal fluids composition in dogs, the present study was aimed to assess the cortisol
concentrations in fetal fluids collected from small-sized purebred newborn puppies born by elective
cesarean section, at term of pregnancy (Meloni et al, 2014). Furthermore we
assessed possible correlations of amniotic and allantoic cortisol concentrations and newborn outcome
at 24 hours of age and with the newborn gender. Fetal fluids cortisol concentrations were also
evaluated for correlation with maternal parity, litter-size, neonatal gender, birth weight and Apgar score
(Veronesi et al, 2009). The study, performed on 50 born alive, normal weighed puppies, without gross
physical malformation, showed that cortisol concentration was higher in allantoic than in amniotic fluid
(p<0.01), even if a strong positive correlation between the two fluids cortisol concentration was found
(p<0.0001; R=0.83). Interestingly, higher amniotic (p<0.05) cortisol concentrations were associated to
puppies not surviving at 24 hours after birth. Therefore it could be suggested that this parameter may
be useful for the recognition, at birth, of puppies needing special surveillance in the first day of age. In
relation to the other evaluated parameters, no correlations with amniotic or allantoic cortisol
concentrations were found. In conclusion, the present results showed that in small-sized purebred
puppies, born at term by elective caesarean section, the evaluation of amniotic cortisol concentration
seems useful for the detection of puppies that need special surveillance during the first 24 hours of age,
and should be coupled to the newborn evaluation by Apgar score. Keywords: cortisol, fetal fluids, pregnancy, dog Published in RUNG: 12.05.2017; Views: 4380; Downloads: 248 Full text (711,48 KB) |

266. Ex Vivo Molecular Rejuvenation Improves the Therapeutic Activity of Senescent Human Cardiac Stem Cells in a Mouse Model of Myocardial InfarctionElisa Avolio, Giuseppe Gianfranceschi, Angela Caragnano, Emmanouil Athanasakis, Rajesh Katare, Daniela Cesselli, Marco Meloni, Anita Palma, Arianna Barchiesi, Carlo Vascotto, Barbara Toffoletto, Elisa Mazzega, Nicoletta Finato, Giuseppe Aresu, Ugolino Livi, Costanza Emanueli, Giacinto Scoles, Carlo Alberto Beltrami, Paolo Madeddu, Antonio Paolo Beltrami, 2014, original scientific article Abstract: Cardiac stem cells (CSC) from explanted decompensated hearts (E-CSC) are, with respect to those obtained from healthy donors (D-CSC), senescent and functionally impaired. We aimed to identify alterations in signaling pathways that are associated with CSC senescence. Additionally, we inves- tigated if pharmacological modulation of altered pathways can reduce CSC senescence in vitro and enhance their reparative ability in vivo. Measurement of secreted factors showed that E-CSC release larger amounts of proinflammatory cytokine IL1b compared with D-CSC. Using blocking antibodies, we verified that IL1b hampers the paracrine protective action of E-CSC on cardiomyo- cyte viability. IL1b acts intracranially inducing IKKb signaling, a mechanism that via nuclear factor-jB upregulates the expression of IL1b itself. Moreover, E-CSC show reduced levels of AMP protein kinase (AMPK) activating phosphorylation. This latter event, together with enhanced IKKb signaling, increases TORC1 activity, thereby impairing the autophagic flux and inhibiting the phos- phorylation of Akt and cAMP response element-binding protein. The combined use of rapamycin and resveratrol enhanced AMPK, thereby restoring downstream signaling and reducing IL1b secretion. These molecular corrections reduced E-CSC senescence, re-establishing their protective activity on cardiomyocytes. Moreover ex vivo treatment with rapamycin and resveratrol improved E-CSC capacity to induce cardiac repair upon injection in the mouse infarcted heart, leading to reduced cardiomyocyte senescence and apoptosis and increased abundance of endog- enous c-Kit1 CSC in the peri-infarct area. Molecular rejuvenation of patient-derived CSC by short pharmacologic conditioning boosts their in vivo reparative abilities. This approach might prove useful for refinement of CSC-based therapies. 270. Donor doping of K0.5Na0.5NbO3 ceramics with strontium and its implications to grain size, phase composition and crystal structureJitka Hreščak, Goran Dražić, Marco Deluca, Iztok Arčon, Alojz Kodre, M. Dapiaggi, Tadej Rojac, Barbara Malič, Andreja Benčan, 2017, original scientific article Abstract: In this study, the particular effects of A-site donor doping in a lead-free piezoceramic material K0.5Na0.5NbO3 (KNN) doped with Sr2+, i.e., the crystal-structure change, the secondary-phase formation and the grain-size decrease, were investigated. The already-reported causes of these effects upon doping KNN were critically discussed and a mechanism of the effects’ formation was suggested and experimentally supported with advanced analytical methods. Extended X-ray absorption fine structure (EXAFS) analyses proved that the Sr occupies the perovskite A-sublattice, and locally modifies the KNN monoclinic structure to cubic. With the help of Sr K-edge EXAFS and wavelength-dispersive X-ray spectroscopy, the Sr was found to be homogenously distributed in the KNN perovskite lattice with 0.5, 1 and 2% Sr and no Sr segregation on the nano level was found in any of the studied samples with transmission electron microscopy. Introducing Sr into the A-sublattice, as well as accounting for the charge-compensating A-site vacancies in the starting composition, causes increasing lattice disorder and microstrain, as determined from a Rietveld refinement of the synchrotron X-ray diffraction data. Above 2% Sr the system segregates the A-site vacancies in a secondary phase in order to release the chemical pressure, as revealed by Raman spectroscopy. All these effects result in an increasing number of low-angle grain boundaries that limit the grain growth and finally lead to a significant grain-size decrease.
