CHARACTERIZATION OF SLOVENIAN APPLE JUICE WITH RESPECT TO ITS GEOGRAPHICAL ORIGIN AND AGRICULTURAL PRODUCTION PRACTICE

Opis: Determination of food authenticity is an important issue in quality control and food safety. Recent studies predict a growing demand for natural and more authentic food and beverage products. The quality and authenticity of apple juice is also of a great economic importance since the popularity and demand for apple juice consumption has increased. The growth of the market for organically produced apples and apple juice is due to the increasing demand for healthy food requirements, protection of the environment and the promotion of biotic diversity. Organic foods have a higher nutritional and health value, but they are more expensive, because their production is more difficult and less profitable. In addition to how food is produced, consumers are increasingly placing emphasis on food products of specific region, which are known for their unique natural flavours and taste. The presented thesis is based on four separate but closely interrelated studies, in which a combination of different isotopic ratios of bioelements (2H/1H, 13C/12C, 15N/14N, 18O/16O), multi-element analysis, and major primary and secondary metabolite profiles were exploited to differentiate the geographical origin and agricultural production practice (organic vs integrated/conventional) of Slovenian apples. These parameters were used to establish the first database of authentic Slovenian apple juice, which can be used to verify the authenticity of commercially available apple juice in Slovenia. The first preliminary study was entitled “Organic Cultivation ~ Geographical Origin (OCGO)” and was performed using apples from the 2009 growing season. Its aim was to examine the use of stable isotope and multi-element data for determining the geographical origin and agricultural production practice of fresh apple juices. Fruits of six apple (Malus domestica Borkh) cultivars (Topaz, Idared, Golden Delicious, Goldrush, Gala, Gloster) were collected from four different geographical regions of Slovenia (Alpine, Dinaric, Pannonian and Mediterranean) grown under organic and integrated/conventional orchard management systems. The results revealed that stable isotope parameters in sugar, pulp and water were the most significant variables for differentiating between the regions. Good separation was achieved between the geographical regions in Slovenia based on the δ18O and δ2H values in water and Rb and S levels in the apple fruit juice. The most significant variables that distinguished between organically and integrated/conventionally cultivated apples were the 15N/14N ratio and antioxidant activity of the apple juice. Significant differences were also observed in the ascorbic acid content of the juice. Based on these results the number and types of apples and the minimum number of samples needed from the same region for determining geographical origin were determined. The second study was called “Organic ~ Conventional Apple Cultivation” (OCAC) and was performed in 2010 and 2011 in a Gala apple orchard. The aim was to determine the effect of different fertilizers allowed either in organic or conventional/integrated agricultural regimes on different parameters. Quality parameters, isotopic composition of C in sugars and in pulp together with N and elemental analysis were investigated. The following five fertilizers were applied: Biosol and Plantella organic (organic) and Ca cyanamide, KAN and UREA (mineral) at a rate of 60 and 120 kg of nitrogen per hectare. From the obtained data it was possible to differentiate between organic and integrated/conventional apple production when taking into account the following parameters: mass, skin and flesh firmness (SFF), total soluble solids (TSS), and the content of Cl as well as δ15N and δ13C in the pulp. PHOTOLYSIS AND ECOTOXICITY OF THE ANTIBIOTIC CIPROFLOXACIN

Opis: Over the last few decades, numerous pharmaceuticals coming from domestic sewage, livestock, hospitals and industries have been determined in ground, surface and drinking waters. A common example of these pollutants are fluoroquinolones, which are powerful antibiotics used in human and veterinary medicine. The presence of fluoroquinolones in the environment even at low concentration constitutes a threat to ecosystems and human health because they can lead to bacterial resistance issues. Up to now the environmental impacts of fluoroquinolones and their degradation products are not fully understood but it is known that they can negatively affect plants and aquatic organisms. In this study the degradation of ciprofloxacin (CIP) present in synthetic freshwater was investigated under simulated sunlight. Concentrations before and after degradation were determined using HPLC-DAD. The mineralisation rate of the treated solution was monitored on a total organic carbon analyser. The ecotoxicity of the initial samples and the degradation mixtures was assessed on organisms belonging to different trophic levels: a marine bacterium (Vibrio fischeri), a freshwater invertebrate (Daphnia magna) and seed clover (Trifolium repens). In parallel, the EC50 of CIP was determined for all organisms. The Vibrio fischeri and Trifolium repens tests showed that EC50 value was more than 250 mg/L and for Daphnia magna was 190 mg/L. Photolysis of CIP showed that after 90 min of irradiation, CIP was almost totally degraded (99%). Photolysis process of CIP led to the formation of toxic by-products towards Vibrio fischeri. The toxicity test results on Trifolium repens did not show significant difference between the control and the treated plants. Thus, CIP and its degradation products have no effect on the germination or root length of Trifolium repens. Tests on Daphnia magna after 240 min irradiation showed a 94% reduction of the toxic effect. Non-purgeable organic carbon (NPOC) analysis showed that there is no significant mineralisation of the sample even after 240 min of irradiation.

Tematika ujetosti v romanu Intimno Gabriele Babnik

Opis: Z literarnovedno analizo in interpretacijo romana Intimno ter na podlagi izsledkov socioloških in migracijskih študij smo ugotavljali, kako se priseljenci soočajo z novo kulturo, s stereotipi in predsodki, ki jih Slovenci izražajo do njih, in ali se priseljencem uspe integrirati in asimilirati v neznano okolje. Preučevali smo ujetost na družbeni in osebno-intimni ravni, ter kako ta vpliva na razmišljanje priseljencev. Preverjali smo odnose med moškim in žensko, odnose med starši in otroki ter kako ti vplivajo na izgradnjo identitete ter kako se ta spreminja in preoblikuje. Ugotovili smo, da se priseljenci soočajo s stereotipi in predsodki, kar jim onemogoča, da bi se uspešno integrirali in asimilirali v novo okolje. Zaradi zavračanja s strani domačinov se počutijo tujci v novem okolju, hkrati pa občutijo tudi odtujitev od domovine. Spopadanje z vsakdanjimi očitki in težavami povzroči izgubo njihove identitete, s tem pa se pojavijo tudi konflikti v partnerskih odnosih. Odnosi med starši in otroki se kažejo, kot slabi, zaradi česar se odrasli posamezniki ne znajo soočati s težavami v partnerskih odnosih in pri vzgoji otrok.

Prenova sistema za spremljanje usposobljenosti zaposlenih v podjetju Mahle Letrika

Opis: V podjetju Mahle Letrika d.o.o. so na področju upravljanja znanja naredili že veliko, vendar so rešitve nepovezane, ponekod nepregledne in z določenimi pomanjkljivostmi. Potrebne so dopolnitve in prenova. Podjetje planira informatizacijo skozi obstoječo matriko usposobljenosti zaposlenih, ki pa jo je potrebno najprej posodobiti in izboljšati njeno funkcionalnost. V podjetju je bila sestavljena skupina sodelavcev za pregled pomanjkljivosti obstoječe matrike in izdelavo načrta sprememb. V prispevku opisujemo pregled obstoječe matrike in njenih pomanjkljivosti ter konkretne zamisli za popravke obstoječega stanja pred nadaljnjo informatizacijo. Rezultati bodo vključeni v načrt aktivnosti za posodabljanje in urejanje integriranega sistema, katerega cilj bo zbiranje informacij o znanju zaposlenih. Zaradi večje sledljivosti se bo izboljšalo upravljanje s človeškimi viri.

Automated procedures for the Fluorescence Detector calibration at the Pierre Auger Observatory

Opis: The quality of the physics results, derived from the analysis of the data collected at the Pierre Auger Observatory depends heavily on the calibration and monitoring of the components of the detectors. It is crucial to maintain a database containing complete information on the absolute calibration of all photomultipliers and their time evolution. The low rate of the physics events implies that the analysis will have to be made over a long period of operation. This requirement imposes a very organized and reliable data storage and data management strategy, in order to guarantee correct data preservation and high data quality. The Fluorescence Detector (FD) consists of 27 telescopes with about 12,000 phototubes which have to be calibrated periodically. A special absolute calibration system is used. It is based on a calibrated light source with a diffusive screen, uniformly illuminating photomultipliers of the camera. This absolute calibration is performed every few years, as its use is not compatible with the operation of the detector. To monitor the stability and the time behavior, another light source system operates every night of data taking. This relative calibration procedure yields more than 2×10[sup]4 raw files each year, about 1 TByte/year. In this paper we describe a new web-interfaced database architecture to manage, store, produce and analyse FD calibration data. It contains the configuration and operating parameters of the detectors at each instant and other relevant functional parameters that are needed for the analysis or to monitor possible instabilities, used for the early discovery of malfunctioning components. Based on over 10 years of operation, we present results on the long term performance of FD and its dependence on environmental variables. We also report on a check of the absolute calibration values by analysing the signals left by stars traversing the FD field of view.

Samostalniški lažni prijatelji v jezikovnem paru slovenščine in srbščine

Opis: Lažni prijatelji so v jezikoslovju imenovani pari besed v različnih jezikih, ki so videti podobno, imajo pa drugačen pomen. Ti besedni pari povzročajo velike probleme pri prevajanju in učenju tujega jezika. Še posebej v sorodnih jezikih, kjer so besede lahko izgovorno popolnoma enake (npr. hlad : hlad), pogosto tudi etimološko sorodne, zato se nam posledično zdijo tudi pomensko povezane, kar pa ni vedno res (hlad v srbščini namreč pomeni senca). Lažnih prijateljev je veliko med slovanskimi jeziki. Jaz sem se odločila za pregled srbskih in slovenskih lažnih prijateljev. V diplomskem delu sem predstavila lažne prijatelje med srbskim in slovenskim jezikom, in sicer samostalnike v imenovalniku ednine, ki so videti popolnoma enaki (npr. lice - lice) oz. se razlikujejo v eni do dveh črkah (npr. kovčeg - kovček). Naloga je sestavljena iz dveh delov, in sicer bom najprej predstavila izhodišča za obravnavo lažnih prijateljih in na podlagi obstoječih virov razložila njihovo tipologijo. V drugem delu pa bom podrobneje predstavila slovensko-srbske lažne prijatelje med samostalniki po abecedi od A do Ž, za vsakega navedla slovarsko razlago (v tabelah) in ponazoritvene stavke pridobljene iz Slovarja slovenskega knjižnega jezika ter Rečnika srpskohrvatskoga književnog jezika. Primeri so prevajani enosmerno, kar pomeni, da sem prevajala samo srbske primere v slovenski jezik, in sicer pravilen in nepravilen prevod obravnavane besede. Ker slovar lažnih prijateljev med srbskim in slovenskim jezikom še ne obstaja, želim z nalogo ljudi, ki se učijo tujega jezika tj. srbščine ali slovenščine, opozoriti na problem lažnega prijateljstva in jim pomagati, da se bodo z njim znali soočati.
